目的 初步探讨镉诱导人胚肾细胞损伤的细胞分子机理。方法 采用MTT法 ,观察镉对人胚肾细胞损伤的影响 ;用流式细胞仪检测镉诱导人胚肾细胞凋亡 ;用分光光度计检测镉对肾细胞SOD活性的影响。结果 几种不同剂量的镉加入细胞作用 2 4h ,4 8h ,72h后均能损伤肾细胞 ,抑制细胞生长 ;镉能诱导人胚肾细胞凋亡。并且能降低肾细胞SOD活性。结论 镉对人胚肾细胞生长有一定的抑制作用 ,能诱导人胚肾细胞凋亡及抑制肾细胞SOD活性。
Objective To investigate the molecular mechanism of cadmium-induced human embryonic kidney cell injury. Methods MTT method was used to observe the effects of cadmium on the injury of human embryonic kidney cells. Cadmium-induced apoptosis in human embryonic kidney cells was detected by flow cytometry. The effect of cadmium on SOD activity in renal cells was detected by spectrophotometer. Results Cadmium could induce the apoptosis of human embryo kidney cells after several different doses of cadmium were added into the cells for 24 hours, 48 hours and 72 hours. And can reduce renal cell SOD activity. Conclusion Cadmium can inhibit the growth of human embryonic kidney cells, induce the apoptosis of human embryonic kidney cells and inhibit the activity of SOD in renal cells.
