目的 探讨可能影响蛇毒蛋白 C激活剂作用的实验条件和该试剂的稳定性。方法 分别采用APTT法和 CBS0 2 .46底物法 ,检测蛇毒蛋白 C激活剂在不同实验条件下的抗凝活性及酰胺酶活性。结果 蛇毒蛋白 C激活剂的最适 p H值为 6.0~ 7.0 ,最适温度为 37~ 40℃ ,肝素、枸橼酸钠等抗凝剂及 Na+ 、K+ 、EDTA等不影响其活性 ,SDS则显著抑制其生物学活性。该试剂溶液剂型在室温下或 4℃中 4w内效价无变化 ,冻干剂型则在 2 y内效价无明显变化。结论 蛇毒蛋白 C激活剂是一种效价较高、稳定性较好、影响因素少的蛋白 C活化试剂
Objective To investigate the experimental conditions and the stability of this reagent that may affect the protein C activator of snake venom. Methods APTT and CBS 02.46 substrate method were used to detect the anticoagulant activity and amidase activity of the protein C activator of the snake venom under different experimental conditions. Results The optimal p H value of protein activator from snake venom was 6.0-7.0 and the optimum temperature was 37-40 ℃. The anticoagulants such as heparin and sodium citrate, Na +, K + Significantly inhibited its biological activity. The reagent solution formulation showed no change in titer within 4 w at room temperature or 4 ° C, whereas the lyophilized dosage form showed no significant change in titer within 2 y. Conclusion The snake venom protein C activator is a protein C activation reagent with higher potency, better stability and less influence factors
