A very sensitive and highly reliable detection system for detecting activated Factor IX (FIXa) has been established. This assay system is based on the cleavage of the fluorogenic substrate by activator X (F X a) produced by F IXa. This assay can be used simultaneously to process a large number of samples because it is based on a format suitable for microtiter plates and it is also easy to routinely screen a large number of samples for automated processing. By virtue of this method, the authors were able to determine the FIXa levels in different therapeutic FIX preparations that are commercially available, such as high purity FIX (HPEIX) and prothrombin complex concentrate (PCC).
