目的:建立清火栀麦片的HPLC指纹图谱,并测定清火栀麦片中栀子苷、穿心莲内酯和脱水穿心莲内酯的含量。方法:采用CAPCELL PAK C18柱(250 mm×4.6 mm,5μm),以乙腈-水为流动相,梯度洗脱,流速为1.0 ml·min~(-1),检测波长为225 nm,柱温为40℃。对13批不同厂家清火栀麦片进行分析,建立清火栀麦片的指纹图谱。结果:在HPLC色谱指纹图谱中,标定了14个共有峰,建立了清火栀麦片HPLC特征指纹图谱共有模式。通过对照品归属法,确定了栀子苷、穿心莲内酯和脱水穿心莲内酯3个化合物。13批清火栀麦片中有9批相似度在0.90以上。3种成分的含量测定线性良好(r>0.999 8,n=6),栀子苷、穿心莲内酯和脱水穿心莲的平均回收率为96.90%,97.55%,97.07%,RSD分别为1.61%,1.45%,1.47%(n=9)。结论:本法简便、准确、重复性好,可以用于清火栀麦片的鉴定与评价,可作为清火栀麦片质量控制的重要科学依据之一。
Objective: To establish HPLC fingerprints of Qinghuizhi Miao tablets and to determine the content of Geniposide, Andrographolide and Dehydroandrographolide in Qinghuizhi Miao tablets. Methods: The mobile phase consisted of acetonitrile and water with gradient elution at a flow rate of 1.0 ml · min -1 using a CAPCELL PAK C18 column (250 mm × 4.6 mm, 5 μm) with a detection wavelength at 225 nm and a column temperature of 40 ° C. Analysis of 13 batches of different manufacturers Qinghuizhi Oatmeal, the establishment of Qinghuizhi cereal fingerprints. Results: In HPLC chromatographic fingerprinting, 14 common peaks were calibrated, and the HPLC fingerprint pattern of Qinghuizhi cereal was established. By reference method, identified three compounds gardenia glycosides, andrographolide and dehydroandrographolide. 13 batch Qinghuizhi cereal tablets in nine batches of similarity above 0.90. The average recoveries of geniposide, andrographolide and dehydroandrographolide were 96.90%, 97.55% and 97.07%, respectively, with RSDs of 1.61% and 1.45, respectively, with good linearity (r> 0.999 8, n = 6) %, 1.47% (n = 9). Conclusion: The method is simple, accurate and reproducible. It can be used for identification and evaluation of Qinghuizhi cereal flakes and can be used as one of the important scientific bases for quality control of Qinghuizhu cereal flakes.
