目的观察丹参水提物对大鼠离体心脏缺血再灌注损伤的保护作用。方法采用大鼠离体心脏灌流技术制备心脏缺血再灌注损伤模型,用不同浓度丹参水提物灌流心脏。观察记录复灌50 min时的心肌功能变化。结果丹参水提物可提高心脏缺血再灌期的左心室舒张压(LVDP)、左心室内压最大上升速率(+dp/dtmax)、左心室内压最大下降速率(-dp/dtmax)水平,差异具有统计学意义(P<0.01),降低左心室舒张末压(LVEDP)、肌酸激酶(CK)和乳酸脱氢酶(LDH)水平,差异具有统计学意义(P<0.01),提高心肌中p-Akt水平,差异具有统计学意义(P<0.01)。结论丹参水提物对缺血再灌注损伤心脏具有保护作用,这一保护作用与它调节磷脂酰肌醇-3-激酶/丝苏氨酸蛋白激酶(PI3K/Akt)信号通路有关。
Objective To observe the protective effect of Salvia miltiorrhiza on isolated rat hearts from ischemia-reperfusion injury. Methods Rat heart perfusion was used to prepare the model of heart ischemia-reperfusion injury and the heart was perfused with different concentrations of Salvia miltiorrhiza extracts. Myocardial function changes were observed at 50 min after reperfusion. Results The aqueous extract of Radix Salviae Miltiorrhizae could improve the left ventricular diastolic pressure (LVDP), the maximum rate of left ventricular pressure (+ dp / dtmax) and the maximum rate of left ventricular pressure (-dp / dtmax) , The difference was statistically significant (P <0.01), LVEDP, CK and LDH levels were decreased, the difference was statistically significant (P <0.01), increased The level of p-Akt in myocardium was significantly different (P <0.01). Conclusion The aqueous extract of Radix Salviae Miltiorrhizae can protect the heart from ischemia-reperfusion injury, which is related to its regulation of PI3K / Akt signaling pathway.
