目的通过观察短波辐射对HBV转基因小鼠树突状细胞(DC)分泌白介素-12(IL-12)及T 淋巴细胞分泌IL-10、IFN-γ的影响,从而探讨短波辐射治疗慢性乙型肝炎的可能相关机制。方法选取 HBV转基因小鼠20只,将其随机分为短波组和空白对照组,另取10只健康小鼠作为正常对照组,其中短波组小鼠给予短波辐射。各组小鼠分别于实验进行10 d后取其脾脏分离树突状细胞,经流式细胞仪检测 DC表型,采用ELISA方法检测DC分泌IL-12以及在刺激混合淋巴细胞反应中T淋巴细胞分泌IL-10、IFN- γ的含量变化情况。结果短波组DC分泌IL-12水平较空白对照组显著增高(P<0.01),与正常对照组比较,差异亦有统计学意义(P<0.01);在混合淋巴细胞反应中,短波辐射能显著抑制T淋巴细胞分泌IL-10 (P<0.01),并同时提高IFN-γ水平(P<0.01)。结论短波辐射能显著上调HBV转基因小鼠DC功能,对 HBV所致慢性乙型肝炎的临床治疗具有潜在应用价值。
Objective To investigate the effects of shortwave radiation on the secretion of IL-12 and IL-10 and IFN-γ secreted by dendritic cells (DCs) in HBV transgenic mice and the effects of shortwave radiation on chronic hepatitis B The possible related mechanism. Methods Twenty HBV transgenic mice were selected and randomly divided into short-wave group and blank control group, and another 10 healthy mice were selected as normal control group. Short-wave radiation was given to short-wave mice. Dendritic cells were isolated from the spleens of DCs, DCs were detected by flow cytometry, IL-12 secretion by DCs and T lymphocytes in mixed lymphocyte reaction Secretion of IL-10, IFN-γ content changes. Results The level of IL-12 secreted by short-wave group was significantly higher than that of the blank control group (P <0.01), and the difference was also statistically significant compared with the normal control group (P <0.01). In the mixed lymphocyte reaction, Short-wave radiation significantly inhibited the secretion of IL-10 by T lymphocytes (P <0.01), and at the same time increased the level of IFN-γ (P <0.01). Conclusion Shortwave radiation can significantly upregulate the DC function of HBV transgenic mice, and has potential value in the clinical treatment of chronic hepatitis B caused by HBV.
