目的研究藿香正气散含药血清对小鼠小肠Cajal间质细胞(ICC)中GLUT2、GLUT4含量的影响。方法采用综合造模法建立小鼠胃肠动力障碍模型;采用Ⅱ型胶原酶消化法体外培养ICC,通过给ICC细胞加入藿香正气散抑制型、稳定型和亢进型三种含药血清,测定GLUT2、GLUT4的含量。结果造模2天抑制型含药血清对GLUT2的影响,与空白抑制组比较,模型抑制组、模型恢复抑制组降低,其中模型给药抑制组升高且高于空白抑制组水平,模型恢复抑制组低于空白抑制组水平(P<0.01);对GLUT4的影响不大(P>0.05)。造模2天稳定型含药血清对GLUT2的影响,与模型稳定组比较,模型恢复稳定组升高(P<0.05)。造模2天稳定型含药血清对GLUT4的影响,与空白稳定组比较,模型稳定组、模型恢复稳定组升高(P<0.05);与模型稳定组比较,空白稳定组、模型给药稳定组降低接近空白稳定水平(P<0.05);造模2天亢进型含药血清对GLUT2的影响,与空白亢进组比较,模型亢进组、模型给药亢进组、模型恢复亢进组皆降低,其中模型给药亢进组降低明显,但是都比空白亢进组低(P<0.01);与模型亢进组比较,空白亢进组升高,模型给药亢进组、模型恢复亢进组降低(P<0.01);对GLUT4的影响不大(P>0.05)。结论藿香正气散抑制型、稳定型和亢进型三种含药血清对小鼠小肠Cajal上的GLUT2、GLUT4含量有影响,其中藿香正气散抑制型和亢进性含药血清对GLUT2影响较大,稳定型含药血清对GLUT4影响较大,藿香正气散可能对小鼠胃肠运动具有双向调节作用及肠推动功能有明显、稳定的促进作用,这可能与GLUT2、GLUT4参与胃肠动力调节有关。
Objective To investigate the effect of Huoxiang Zhengqi Powder on the content of GLUT2 and GLUT4 in small intestinal Cajal interstitial cells (ICC) in mice. Methods The model of gastrointestinal motility disorder was established by synthetic modeling. The ICC was cultured in vitro by collagenase digestion method Ⅱ. The ICC cells were treated with Huoxiang Zhengqi San inhibitory, stable and hyperthyroid serums, GLUT2, GLUT4 content. Results Compared with blank control group, the inhibition of GLUT2 by model drug-containing serum for 2 days was lower in model control group and model recovery inhibition group, in which model administration inhibited group was higher than that in blank control group, Group was lower than that of blank control group (P <0.01), and had little effect on GLUT4 (P> 0.05). The effect of 2-day stable drug-containing serum on GLUT2 expression in model stable group was higher than that in model stable group (P <0.05). Compared with the blank control group, the stable model group and the stable model group recovered (P <0.05). Compared with the model control group, the stability of the blank control group and the model administration were stable (P <0.05). The effect of model 2-day hyperthyroidism-containing serum on GLUT2 was significantly lower than that in model group with hyperthyroidism, model hyperthyroidism and model recovery hyperthyroidism Compared with hyperthyroidism group, the hyperthyroidism group was higher than that of hyperthyroidism group (P <0.01), while the hyperthyroidism model group was hyperthyroidism model and hyperthyroidism model group was decreased (P <0.01). Little effect on GLUT4 (P> 0.05). Conclusions Huoxiangzhengqisan inhibitory, stable and hyperthyroid serums have effects on GLUT2 and GLUT4 contents in small intestine Cajal of mice, and the inhibitory and hyperthyroid serums of Huoxiangzhengqisan and hyperthyroidism have a significant effect on GLUT2 , Stable drug-containing serum had a significant effect on GLUT4. Huoxiangzhengqisan could significantly and steadily promote gastrointestinal motility and promote intestinal motility in mice, which may be related to the involvement of GLUT2 and GLUT4 in the regulation of gastrointestinal motility related.
