利用测色氨酸的凝胶过滤色层法对籽粒进行碱水解分析,测定正常玉米(+)和奥帕克2号玉米(02)正在发育籽粒中蛋白质色氨酸的积累作用。由一线性关系式表示籽粒中结合入蛋白质的色氨酸变化同蛋白质总量变化之间的关系。虽然单粒色氨酸含量随籽粒的发育而增加,但它在两种玉米籽粒蛋白质总量中的比例均有所下降,从最初的1.3%分别降到极限值0.55%(+)和0.93%(02)。当籽粒蛋白质含量从正常玉米提高至奥帕克2号的水平时,含色氨酸的蛋白质含量也随之增加。但是,在含色氨酸不多的玉米中,由于蛋白质增加的幅度更大,导致整个籽粒蛋白质中色氨酸的稀释,从而降低了其营养品质。
Alkaline hydrolysis of the seeds was carried out using a tryptophan gel filtration chromatography to determine the accumulation of protein tryptophan in developing kernels of normal maize (+) and Opaque 2 maize (02). The relationship between the change in tryptophan bound to protein in a grain and the change in total protein is expressed by a linear relationship. Although the content of mono-tryptophan increased with the development of grain, its proportion of total protein in both maize grains decreased from the initial 1.3% to the limits of 0.55% (+) and 0.93%, respectively (02). As the grain protein content increases from normal corn to Opaque 2 levels, the tryptophan-containing protein content also increases. However, in maize containing little tryptophan, the protein is more greatly amplified, resulting in the dilution of tryptophan in the entire grain protein, thereby reducing its nutritional quality.
