目的观察碘缺乏和甲状腺功能减退对大鼠仔鼠海马细胞外信号调节蛋白激酶1及2(ERK1/2)表达的影响。方法健康2月龄孕Wistar大鼠28只,按体重随机分成对照组、甲状腺功能减退组[按饮水中含丙基硫尿嘧啶(PTU)剂量分为5mg/L组和15mg/L组]和碘缺乏组,每组7只。分别于出生后第7、14、21、28和42天每组随机取5只仔鼠,灌流固定大脑,用组织病理切片和免疫组化染色观察分析海马的ERK1/2表达。结果在出生后14、21、28和42天时,海马CA1和CA3区的ERK1/2表达在PTU5mg/L组、PTU15mg/L组和碘缺乏组显著低于对照组(P<0.05)。DG区的ERK1/2表达与对照组相比差异无显著性。出生后7天时,各组间ERK1/2表达差异无显著性。结论碘缺乏和甲状腺功能减退可降低海马CA1和CA3区的ERK1/2表达。
Objective To investigate the effects of iodine deficiency and hypothyroidism on the expression of extracellular signal-regulated protein kinases 1 and 2 (ERK1 / 2) in hippocampus of offspring rats. Methods Twenty-eight healthy Wistar rats of 2 months old were randomly divided into control group, hypothyroidism group [divided into 5mg / L group and 15mg / L group according to the concentration of propylthiouracil (PTU) in drinking water) and Iodine deficiency group, 7 rats in each group. At the 7th, 14th, 21st, 28th and 42nd days after birth, 5 rats were randomly selected from each group. Peripheral blood was infused into the brains. The expression of ERK1 / 2 in the hippocampus was analyzed by histopathological sections and immunohistochemical staining. Results The expression of ERK1 / 2 in hippocampal CA1 and CA3 was significantly lower in PTU 5mg / L group, PTU 15mg / L group and iodine deficiency group than those in control group at 14,21,28 and 42 days after birth (P <0.05). The expression of ERK1 / 2 in DG area was not significantly different from the control group. At 7 days after birth, there was no significant difference in ERK1 / 2 expression among groups. Conclusions Iodine deficiency and hypothyroidism can reduce the expression of ERK1 / 2 in CA1 and CA3 of hippocampus.
