目的:建立HPLC同时测定天山花揪中绿原酸、芦丁和金丝桃苷3种成分含量的方法。方法:用60%的乙醇超声提取,Waters C18(250 mm×4.6 mm,5μm)色谱柱,以甲醇-乙腈-0.3%磷酸水为流动相进行梯度洗脱,检测波长360 nm,柱温25℃。结果:绿原酸、芦丁和金丝桃苷分别在33.12~115.9,1.051~11.56,0.8040~8.844μg/mL浓度范围内呈良好的线性关系,相关系数均为0.9999;平均加样回收率(n=3)分别为98.87%、100.2%、101.0%。经测定天山花揪叶中绿原酸、芦丁和金丝桃苷含量分别为1.271%、0.08884%、0.1430%。结论:本法一次性同时测定天山花揪中3种成分含量,方法简便、准确、可靠,可为药材和制剂研究提供较为全面的科学依据及有效的质量控制方法。
Objective: To establish a HPLC method for the simultaneous determination of three components of chlorogenic acid, rutin and hyperoside in T. tianshanensis. Methods: Ultrasonic extraction with 60% ethanol, Waters C18 (250 mm×4.6 mm, 5 μm) column, gradient elution with methanol-acetonitrile-0.3% phosphoric acid as mobile phase, detection wavelength 360 nm, column temperature 25°C . Results: The chlorogenic acid, rutin and hyperoside showed a good linear relationship within the concentration ranges of 33.12-115.9,1.051-11.56, and 0.8040-8.844 g/mL, respectively. The correlation coefficient was 0.9999; average recovery ( n=3) were 98.87%, 100.2%, and 101.0%, respectively. The contents of chlorogenic acid, rutin and hyperoside in the leaves of T. tianshanensis were determined to be 1.271%, 0.08884%, and 0.1430%, respectively. Conclusion: This method can be used to determine the contents of three components in a single-time, simultaneous determination of the components of T. tianshanensis. The method is simple, accurate, and reliable. It can provide a comprehensive scientific basis and effective quality control methods for the study of medicinal materials and preparations.
