目的运用代谢组学方法研究淫羊藿大极性黄酮(EHPF)对正常生理状态大鼠尿液中内源性物质代谢的影响。方法 SD大鼠随机分为空白组、EHPF组,每组10只,EHPF组灌胃给予EHPF提取物21.6 mg·kg~(-1),空白组给予等体积蒸馏水,每日1次,连续20 d,于末次给药后代谢笼收集大鼠12 h尿液,处理后采用超高效液相色谱-飞行时间质谱联用(UPLC-TOF/MS)分析,所得结果进行多元数据分析、潜在生物标志物鉴定及生物学意义分析。结果鉴定出6个潜在生物标志物,归属5条主要代谢通路,EHPF可使5-羟吲哚乙醛和5-甲氧基吲哚乙酸表达升高,促进色氨酸及5-羟色胺代谢途径;还可降低肾上腺素和L-甲状腺原氨酸水平,可能对神经系统、心血管系统、免疫系统具有一定的保护作用;同时EHPF对肉豆蔻酸和三丁酸甘油酯有明显的降低作用,也表现出可能不利于发生炎症时的保护作用及预防肿瘤疾病的潜在毒性。结论 EHPF对正常生理状态大鼠的神经系统、心血管系统、免疫系统具有潜在干预作用,为淫羊藿黄酮类化合物的安全使用及其治疗潜能的开发提供了一定依据。
Objective To investigate the effect of epimedium-polaroid flavonoids (EHPF) on the metabolism of endogenous substances in the urine of normal physiological state rats using metabonomics. Methods Sprague-Dawley rats were randomly divided into blank group and EHPF group with 10 rats in each group. EHPF group was given intragastric administration of EHPF extract 21.6 mg·kg -1 . The blank group was given equal volume of distilled water once a day for 20 consecutive days. d, 12 h urine was collected from metabolic cages after the last administration and analyzed by Ultra Performance Liquid Chromatography-Flight Time-Mass Spectrometry (UPLC-TOF/MS). The results obtained were used for multivariate data analysis and potential biomarkers. Identification and biological significance analysis. The results identified six potential biomarkers, belonging to five major metabolic pathways, EHPF can increase the expression of 5-hydroxy indole acetaldehyde and 5-methoxy indole acetic acid, and promote tryptophan and serotonin metabolism Can also reduce the adrenaline and L-thyronine levels, may have a protective effect on the nervous system, cardiovascular system, immune system; at the same time EHPF on myristic acid and tributyrin have a significant reduction, It also exhibits protective effects that may be detrimental to the occurrence of inflammation and the potential toxicity of preventing tumor diseases. Conclusion EHPF has a potential intervention effect on the nervous system, cardiovascular system and immune system of rats in normal physiological state, which provides a certain basis for the safe use of icari flavonoids and the development of therapeutic potential.
