为探讨Vγ9链T细胞受体(Tcell receptor,TCR)互补决定区3(complementarity determining region3,CDR3)是否参与自身免疫性疾病中γδT细胞的抗原识别过程,利用基因扫描技术和随机序列测定法对系统性红斑狼疮(systemic lupus er-ythematosus,SLE)患者和健康人外周血Vγ9TCR CDR3区的结构特征进行分析。结果显示,Vγ9CDR3序列的变化主要集中在J区等位基因的取用不同:健康人以表达JγP为主(82.86%),而SLE患者Jγ1/Jγ2(40.82%)和JγP(59.18%)表达率相近。两组人群相比,Jγ1/Jγ2和JγP表达率的差异均具有显著性。结果表明,SLE患者外周血Vγ9链T细胞受体具有与健康人不同的CDR3组成特点,提示其可能参与SLE病理过程中γδT细胞的抗原识别。本研究为寻找SLE中γδT细胞识别的自身抗原以及阐明γδT细胞在SLE中的独特作用奠定了基础。
To investigate whether CDR3 of Vγ9 chain T cell receptor (TCR) is involved in the antigen recognition of γδT cells in autoimmune diseases, we used gene-scanning technique and random sequence determination The structural features of Vγ9 TCR CDR3 region in peripheral blood of patients with systemic lupus erythematosus (SLE) and healthy individuals were analyzed. The results showed that the changes of Vγ9 CDR3 sequences were mainly concentrated on the access of alleles in J region: the expression of JγP was predominant in healthy subjects (82.86%), while the expression of Jγ1 / Jγ2 (40.82%) and JγP (59.18%) in SLE patients similar. Compared with the two groups, the difference of Jγ1 / Jγ2 and JγP expression rates were significant. The results showed that peripheral blood Vγ9 chain T cell receptors in SLE patients had different CDR3 composition characteristics from healthy individuals, suggesting that they may be involved in the antigen recognition of γδT cells during SLE pathology. This study laid the foundation for finding the autoantigens recognized by γδT cells in SLE and elucidating the unique role of γδT cells in SLE.
