本研究根据转录组信息从甘蔗细茎野生种(Saccharum Spontaneum)中克隆得到一个抗旱相关的NAC家族基因,命名为Sc NAC1(NCBI accession KP742345)。研究结果显示:该基因包含一个1 212个碱基的开放阅读框,编码403个氨基酸;通过序列比对发现该基因在N端具有保守的NAC结构域,进化树分析显示Sc NAC1属于NAC家族基因的NAP亚家族;通过将Sc NAC1-GFP融合载体转化拟南芥原生质体中,证明Sc NAC1定位于细胞核内;定量PCR检测在甘蔗细茎野生种发育不同时期Sc NAC1对于干旱的响应,发现Sc NAC1在各个时期都受干旱诱导。研究结果可为进一步研究甘蔗干旱响应机理提供参考。
In this study, a drought-resistant NAC family gene was cloned from Saccharum Spontaneum according to the transcriptome information and named Sc NAC1 (NCBI accession KP742345). The results showed that the gene contains an open reading frame of 1212 bp and encodes a protein of 403 amino acids. The sequence of the gene contains a conserved NAC domain at the N-terminus. Phylogenetic analysis showed that Sc NAC1 belongs to the NAC family of genes Sc NAC1-GFP fusion vector was transformed into Arabidopsis thaliana protoplasts, and Sc NAC1 was localized in the nucleus. Quantitative PCR was used to detect the response of Sc NAC1 to drought in different stages of sugarcane stem-stem wild-type development. Sc NAC1 is induced by drought at all times. The results can provide a reference for further research on drought response mechanism of sugarcane.
