目的探讨Foxp3在大鼠骨髓干细胞输注对肾移植免疫耐受的诱导作用及机制。方法大鼠肾移植:40只Wistar大鼠(雌雄各半)随机分为实验组和对照组,进行肾移植手术,术前经门静脉分别注射骨髓细胞及生理盐水,30天后观察肉眼和光镜下排斥反应及应用流式细胞术(FCM)的方法检测T细胞内的Foxp3。结果实验组肉眼和光镜下排斥反应均较轻微,Foxp3在T细胞内Foxp3的表达量明显增加。结论应用他克莫司并输注供体骨髓干细胞,可诱导一定的免疫耐受,此免疫耐受可能与Foxp3的表达上调有关。
Objective To investigate the induction and mechanism of Foxp3 transfusion in rat bone marrow stem cells on immune tolerance of renal allografts. Methods Rat renal transplantation: 40 Wistar rats (male and female, half) were randomly divided into experimental group and control group for renal transplantation. Bone marrow cells and normal saline were injected through the portal vein preoperatively. After 30 days, macroscopic and light microscope exclusion Response and application of flow cytometry (FCM) method to detect Foxp3 T cells. Results The rejection in both eyes and light microscopy in the experimental group was slight, and the Foxp3 expression in T cells was significantly increased. Conclusion The application of tacrolimus and infusion of donor bone marrow stem cells can induce certain immune tolerance, which may be related to the upregulation of Foxp3 expression.
