目的:观察大黄素对动脉损伤后离体培养的平滑肌细胞增殖细胞核抗原(proliferatingcelnuclearantigen,PCNA)和细胞周期时相的影响,了解大黄素抑制离体培养的平滑肌细胞增殖的作用机制。方法:指数增长期的平滑肌细胞同步化于G0期后,加入20mg·L-1大黄素于含胎牛血清10%(体积分数)的细胞培养液中,24h后分别用流式细胞仪进行细胞周期时相测定和抗PCNA蛋白单克隆抗体染色。结果:与对照组比较,用了大黄素以后,G0/G1期细胞百分比升高,S期细胞百分比下降,下降率为75.5%,平滑肌细胞抗PCNA单克隆抗体染色呈阴性。表明PCNA蛋白的表达受到抑制,细胞受阻于G0期。结论:抑制PCNA蛋白表达、阻滞细胞周期的移行是大黄素抑制平滑肌细胞增殖的重要机制之一。
OBJECTIVE: To observe the effect of emodin on proliferation proliferating cell nuclear antigen (PCNA) and cell cycle phase in vitro after injury of arterial injury and to investigate the mechanism of emodin inhibiting the proliferation of smooth muscle cells in vitro. Methods: Synchronization of exponential phase of smooth muscle cells in G0 phase, adding 20mg · L-1 emodin in fetal bovine serum containing 10% (volume fraction) of cell culture medium, respectively, after 24 hours by flow cytometry cells Periodic phase determination and anti-PCNA monoclonal antibody staining. Results: Compared with the control group, the percentage of cells in G0 / G1 phase was increased and the percentage of cells in S phase was decreased with emodin, the decreasing rate was 75.5%. The anti-PCNA monoclonal antibody staining of smooth muscle cells was negative. The results showed that the expression of PCNA protein was inhibited and the cells were blocked in G0 phase. CONCLUSION: Inhibition of PCNA protein expression and cell cycle arrest is one of the important mechanisms by which emodin inhibits the proliferation of smooth muscle cells.
