羟基喜树碱柔性脂质体冻干剂抗肿瘤药效学研究

目的研究羟基喜树碱柔性脂质体冻干剂体内外抗肿瘤作用。方法以市售羟基喜树碱注射液为对照,采用MTT法分别进行了羟基喜树碱柔性脂质体冻干剂对肝癌Hep G-2细胞、胃癌BGC-823细胞及结肠癌LOVO细胞体外抗肿瘤作用,并以人肝癌Hep G-2细胞建立了荷瘤裸鼠动物模型,考察了羟基喜树碱柔性脂质体冻干剂的体内抗肿瘤作用。结果羟基喜树碱柔性脂质体冻干剂对肝癌Hep G-2、胃癌BGC-823具有抗肿瘤作用且效果优于市售羟基喜树碱注射液,但对结肠癌LOVO细胞具有体外抗肿瘤作用较市售羟基喜树碱注射液要差,对裸鼠肝癌Hpe G-2移植瘤的生长有较好的抑制作用且作用强于阳性对照药市售羟基基喜树碱。结论柔性脂质体冻干剂后在一定程度上提高了羟基喜树碱的抗肿瘤作用。 Objective To study the antitumor effect of hydroxycamptothecin flexible liposomes lyophilized in vitro and in vivo. METHODS: Commercially available hydroxycamptothecin injection was used as a control. MTT assay was performed in vitro against HPC-Hep G-2 cells, gastric cancer BGC-823 cells and colon cancer LOVO cells, respectively. Tumor effect, and human hepatoma Hep G-2 cells were established animal model of tumor-bearing nude mice and investigated the anti-tumor effect of hydroxycamptothecin flexible liposome lyophilized agent in vivo. Results Hydroxycamptothecin flexible liposomes lyophilized agent had anti-tumor effect on HepG-2 and BGC-823, and had better anti-tumor effect than the commercial hydroxycamptothecin injection, but had the anti-tumor effect on colon cancer LOVO cells in vitro The effect was worse than that of the commercial hydroxycamptothecin injection and had a better inhibitory effect on the growth of HpG-2 xenografts in nude mice and the effect was stronger than that of the hydroxycamptothecin sold as positive control. Conclusions Flexible liposomes lyophilized to a certain extent, enhance the anti-tumor effect of hydroxycamptothecin.
