目的:通过桃仁涂膜剂用于移植人体肥厚性瘢痕裸鼠模型,以观察瘢痕内微循环、相关因子、自由基等多层次靶点抑制瘢痕作用,探讨桃仁活血化瘀中药抗肥厚性瘢痕的作用机制,为临床中医预防和治疗瘢痕用药提供实验依据。方法:制作桃仁涂膜剂,用于移植人体肥厚性瘢痕裸鼠模型,用游标卡尺测量瘢痕大小,M asson,s多纤维染色法观察分析瘢痕组织切片染色形态,EnV ision法免疫组化检测TGF-β1和CD44含量表达,样本碱水解法生化检测Hyp含量和SOD活力,微循环仪检测瘢痕内微循环。结果:桃仁涂膜剂可促使瘢痕组织内胶原纤维排列疏松并趋于整齐,改善瘢痕内微循环,促使微血管生成;使瘢痕组织内TGF-β1和CD44含量表达下调;使瘢痕组织内Hyp含量明显下降;可提高瘢痕组织内SOD的活力。结论:桃仁活血化瘀中药具有改善瘢痕内微循环,调控相关因子,清除自由基等多层次靶点抑制瘢痕的作用。
OBJECTIVE: To observe the effect of microcirculation, related factors, free radicals and other multi-level targets on the scar inhibition by means of peach kernel-coating agent in transplanted human hypertrophic scar model in nude mice and to study the anti-hypertrophic scars of peach kernel promoting blood circulation and removing blood stasis Mechanism of action for clinical prevention and treatment of traditional Chinese medicine to provide experimental evidence of scar medication. Methods: The peach kernel film was prepared and used to transplan the model of hypertrophic scar in nude mice. The scar size was measured by vernier caliper. The morphology of scar tissue was observed by M on s, s multi-fiber staining. EnV ision method was used to detect the expression of TGF- The expression of β1 and CD44 were detected. Hyp content and SOD activity were detected by alkali hydrolysis and microcirculation. Results: Peach kernel coating could promote collagen fibers in scar tissue to become loose and neatly arranged, improve microcirculation in scar and promote angiogenesis, reduce the expression of TGF-β1 and CD44 in scar tissue, and significantly increase the content of Hyp in scar tissue Decline; can improve SOD activity in scar tissue. Conclusion: Chinese herb of peach and blood circulation promoting blood circulation can improve scarring microcirculation, regulate related factors and eliminate free radicals and other multi-level targets to inhibit scarring.
