目的:探讨中药复方制剂仙珍骨宝对糖皮质激素致大鼠骨质疏松的影响,并比较量效关系。方法:36只3月龄♂SD大鼠,随机分为激素模型(灌胃醋酸泼尼松4.5mg·kg-1,2次/周)和仙珍骨宝高、中、低剂量组(0.2,0.4,0.8g·kg-1·d-1ig6次/周),实验于第90天处死,取胫骨上段行不脱钙骨制片骨形态计量学分析。结果:激素模型组较年龄对照组骨量减少,破骨细胞增多,骨形成参数减少,仙珍骨宝组与糖皮质激素模型对照,明显抑制骨吸收,且骨形成参数增加,阻止骨量的丢失(%Tb.Ar+91.23%),高剂量有明显疗效,中低剂量无效。结论:仙珍骨宝胶囊有预防糖皮质激素引起大鼠骨丢失的作用,高剂量疗效显著。
OBJECTIVE: To investigate the effect of Xianzhenggubao, a traditional Chinese medicine compound preparation, on glucocorticoid-induced osteoporosis in rats and to compare the dose-effect relationship. METHODS: Thirty-six 3-month-old Sprague-Dawley rats were randomly divided into hormone model (4.5 mg·kg-1 prednisone acetate 2 times per week) and Xianzhenggu high, middle and low dose groups (0.2 ,0.4,0.8g·kg-1·d-1ig6 times/week). The experiment was performed on the 90th day. The humerus bone slices were taken for the decalcified bone preparation for morphometric analysis. RESULTS: Compared with the age control group, the hormone model group had less bone mass, more osteoclasts, and less bone formation parameters. Xianzhen Gubao group and the glucocorticoid model were significantly inhibited in bone resorption, and the bone formation parameters increased, preventing bone mass. Lost (%Tb.Ar+91.23%), high dose has obvious curative effect, middle and low doses are invalid. Conclusion: Xianzhen Gubao capsule can prevent glucocorticoid-induced bone loss in rats, and the effect of high dose is significant.
