本文用反相高效液相色谱法测定大鼠静脉注射盐酸小檗胺后,不同时间的血药浓度。血样经乙腈去蛋白后,以乙腈-水(28∶72)为流动相(pH=1.5),流动相流速为1ml/min,紫外检测波长为282nm,血浆中盐酸小檗胺的最低检出极限为10ng,平均回收率为98.6±3.4%。在所选定的色谱条件下,盐酸小檗胺的保留时间为5.6min。
In this study, RP-HPLC was used to determine plasma concentration of different time after intravenous injection of berbamine hydrochloride in rats. After the blood sample was deproteinized by acetonitrile, the mobile phase consisted of acetonitrile and water (pH = 1.5) with mobile phase of 1ml / min and UV detection wavelength of 282nm. The lowest level of berbamine in plasma The limit was 10 ng and the average recovery was 98.6 ± 3.4%. At the selected chromatographic conditions, the retention time of berbamine hydrochloride was 5.6 min.
