目的 探讨鼻咽癌放疗前及放疗中细胞凋亡的变化及其意义。方法 采用TdT酶介导的生物素化dutp缺口末端标记技术 (TUNEL) ,检测 35例鼻咽癌患者放疗前及放疗中鼻咽部刷落癌细胞的细胞凋亡率 (apoptosisrate ,AR)。结果 鼻咽癌放疗前的AR为 7 2 %± 2 4 % ;放疗量分别为 10GY、30GY和 60GY时的AR分别为 64 3%± 2 1 4 %、57 2 %± 17 3%、16 2 %± 7 4 % ,明显高于放疗前 (P <0 0 1) ;放疗前的自发性细胞凋亡与放射治疗诱发的细胞凋亡呈正相关。结论 检测鼻咽癌患者刷落癌细胞的细胞凋亡率有助于判断放射敏感性。
Objective To investigate the changes of apoptosis in nasopharyngeal carcinoma before and during radiotherapy and its significance. Methods TdT-mediated biotinylated dutp nick end labeling (TUNEL) was used to detect the apoptotic rate (AR) of 35 nasopharyngeal carcinoma patients before and during radiotherapy. Results The AR before radiotherapy of nasopharyngeal carcinoma was 72% ± 24%. The AR at 10GY, 30GY and 60GY were 64 3% ± 2 1 4%, 57 2% ± 17 3% and 16 2 respectively % ± 7.4%, which was significantly higher than that before radiotherapy (P <0.01). There was a positive correlation between spontaneous apoptosis before radiotherapy and apoptosis induced by radiotherapy. Conclusion Detecting nasopharyngeal carcinoma cells in the rate of apoptosis of brushing cancer cells help to determine radiosensitivity.
