目的 探讨凋亡相关基因Bax的表达水平与肺癌发生、发展及预后的关系。方法 应用免疫组化链亲和素—生物素标记法 (LSAB法 )检测 12 4例人非小细胞肺癌组织中Bax基因产物的表达水平。结果 肺癌组织中Bax基因产物的表达水平 (5 3 .76 % )明显低于癌旁肺组织 (75 .5 1% )和正常肺组织 (74.97% ) (P <0 .0 1)。肺癌组织中Bax基因产物的表达水平与肺癌P TNM分期、原发肿瘤大小及淋巴结转移状态有密切关系 (P <0 .0 1或P <0 .0 5 ) ,而与肺癌组织学类型、细胞分化程度、原发肿瘤部位、患者的性别、年龄及吸烟与否均无明显关系 (P >0 .0 5 )。肺癌患者Bax基因产物高表达组 5年生存率(49.99% )明显高于低表达组 (11.48% ) (P <0 .0 1)。结论 Bax基因表达异常可能在肺癌发生、发展和预后中起重要作用。Bax基因可能参与了肺癌发生、发展和转移过程
Objective To investigate the relationship between the expression of apoptosis-related gene Bax and the occurrence, development and prognosis of lung cancer. Methods Immunohistochemical streptavidin-biotin labeling (LSAB method) was used to detect the expression of Bax gene product in 124 cases of human non-small cell lung cancer. Results The expression level of Bax gene product in lung cancer tissues (53.76 %) was significantly lower than that in paratumor lung tissues (75.51 %) and normal lung tissues (74.97%) (P < 0.01). The expression of Bax gene product in lung cancer tissues was closely related to the P TNM stage, primary tumor size and lymph node metastasis status (P < 0.01 or P <0.05), but with histological types and cells of lung cancer. There was no significant relationship between the degree of differentiation, the location of the primary tumor, the patient’s gender, age, and smoking (P > 0.05). The 5-year survival rate (49.99%) in patients with high expression of Bax gene products in lung cancer patients was significantly higher than that in the low-expression group (11.48%) (P < 0.01). Conclusion Abnormal expression of Bax gene may play an important role in the occurrence, development and prognosis of lung cancer. Bax gene may be involved in the occurrence, development and metastasis of lung cancer
