为研究 5 -氟脲嘧啶 (5 - FU)对胆囊癌细胞凋亡的影响 ,在体外培养的胆囊癌细胞中加入不同浓度的 5 -FU ,应用光镜、电镜、DNA凝胶电泳及流式细胞仪检测癌细胞凋亡的形态学特征、生化学特征、细胞凋亡百分率及细胞周期变化。在 5 - FU作用下 ,凋亡的胆囊癌细胞出现膜小泡、凋亡小体等特征性改变 ;DNA电泳呈现典型的“梯状”条带 ;流式细胞仪测定出现典型的凋亡峰 ;其凋亡百分率随着药物浓度的提高而明显升高 (与对照组比较 ,P<0 .0 5 )。认为 5 - FU可诱导胆囊癌细胞发生凋亡 ,其凋亡率随药物浓度的提高而升高
To study the effect of 5-fluorouracil (5-FU) on apoptosis of gallbladder cancer cells, different concentrations of 5-FU were added to gallbladder cancer cells cultured in vitro. Light microscope, electron microscope, DNA gel electrophoresis and flow cytometry were applied. Cytological features, biochemical characteristics, percentage of apoptosis, and cell cycle changes of cancer cells were detected by cytometry. Apoptotic gallbladder cancer cells showed characteristic changes of membrane vesicles and apoptotic bodies under the action of 5-FU; DNA electrophoresis showed a typical “ladder” band; flow cytometry showed typical apoptotic peaks. The percentage of apoptotic cells increased significantly with increasing drug concentration (P<0.05 compared with control group). It is believed that 5-FU can induce apoptosis of gallbladder cancer cells, and its apoptosis rate increases with increasing drug concentration.
