建立了血中华法林对映体的毛细管电泳分析方法。电泳缓冲液由140mmol/L磷酸盐缓冲液和7mmol/L的甲基β-环糊精组成(pH7.9)。工作电压13kV,检测波长210nm。两种对映体S(-)-华法林及R(+)-华法林达到基线分离,在0~5μg/ml范围内呈线性。血中华法林对映体最低检测浓度均为0.02μg/ml,精密度及回收率均较好。
A capillary electrophoresis method was developed for the determination of warfarin enantiomers in blood. The electrophoresis buffer consisted of 140 mmol / L phosphate buffer and 7 mmol / L methyl β-cyclodextrin (pH 7.9). Working voltage 13kV, detection wavelength 210nm. The two enantiomers S (-) - warfarin and R (+) - warfarin baseline separation, in the 0 ~ 5μg / ml range was linear. The minimum detectable concentration of warfarin enantiomer in blood was 0.02μg / ml, and the precision and recovery rate were good.
