目的:探讨结肠Cajal间质细胞(ICC)、干细胞因子(SCF)在溃疡性结肠炎(UC)发病中的作用。方法:将BALB/c小鼠随机分为正常对照组(n=8)和UC组(n=24),于造模后1、4、7天分别处死UC组8只小鼠,取远端结肠,用Westernblot方法检测SCF的表达;免疫组化检测结肠ICC的变化,并进行结肠组织学评分。结果:结肠组织学评分在7天达最高值(P<0.05)。与正常小鼠比较,UC小鼠结肠肌间ICC表达明显减少(P<0.05),结肠SCF的表达也显著减少(P<0.01),SCF与ICC呈正相关(UC4天时,r=0.862,P=0.027;UC7天时,r=0.948,P=0.004)。结论:UC小鼠远端结肠组织内SCF表达下降,伴随ICC数量减少,提示UC病变可能与SCF/Kit系统下调有一定的相关性。
Objective: To investigate the role of colon Cajal interstitial cells (ICC) and stem cell factor (SCF) in the pathogenesis of ulcerative colitis (UC). Methods: BALB / c mice were randomly divided into normal control group (n = 8) and UC group (n = 24). Eight mice in UC group were sacrificed at 1, Colon, and the expression of SCF was detected by Western blot. The changes of ICC in colonic mucosa were detected by immunohistochemistry, and the histological score of colon was also evaluated. Results: The colon histological score reached the highest value on the 7th day (P <0.05). Compared with normal mice, the expression of ICC in colonic muscle of UC mice decreased significantly (P <0.05), and the expression of SCF in colon decreased significantly (P <0.01). There was a positive correlation between SCF and ICC (UC = 0.862, P = 0.027; r = 0.948 at day 7 of UC, p = 0.004). CONCLUSIONS: The expression of SCF in distal colon of UC mice decreased with the decrease of ICC, suggesting that UC may be related to the down-regulation of SCF / Kit system.
