目的研究分析ICAM-1K 469E基因多态性与汉族人脑梗死形成的关系。方法选取2009年7月—2013年10月汉族住院脑梗死患者213例,采用聚合酶链反应和限制性片段长度多态性分析(PCR-RFLP)检测确定基因型。对照组224例,均为同期汉族健康体检者,年龄、性别与脑梗死组相匹配,各试验指标检测同脑梗死组。结果脑梗死不同面积与ICAM-1指标关系,脑梗死组(大面积)的ICAM-1指标明显高于脑梗死组(小面积)和脑梗死组(腔隙性),比较差异具有统计学意义(P<0.05);脑梗死组(小面积)和脑梗死组(腔隙性)的ICAM-1指标差异无统计学意义(P>0.05);2组基因型、等位基因分布比较,ICAM-1基因型中,脑梗死组KK基因型显著多于对照组,EE基因型略低于对照组,比较差异具有统计学意义(P<0.05);2组EK基因型比较差异无统计学意义(P>0.05);2组等位基因型分布比较,K/E脑梗死组显著高于对照组,比较差异具有统计学意义(P<0.05)。结论汉族人脑梗死患者与ICAM-1K469E基因多态性存在明确相关性,并且脑梗死面积的不同也与ICAM-1指标相关联,E等位基因可能是影响汉族人脑梗死的重要遗传基因。
Objective To study the relationship between ICAM-1K 469E gene polymorphism and cerebral infarction in Han nationality. Methods 213 patients with cerebral infarction in hospitalized Han nationality from July 2009 to October 2013 were selected and genotypes were determined by polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP). The control group of 224 cases, all the same period, Han healthy subjects, age, gender and cerebral infarction matched, each test indicators with cerebral infarction group. Results The relationship between ICAM-1 and different areas of cerebral infarction was significant. ICAM-1 was significantly higher in cerebral infarction group (large area) than that in cerebral infarction group (small area) and cerebral infarction group (lacunar), the difference was statistically significant (P <0.05). There was no significant difference in ICAM-1 between cerebral infarction (small area) and cerebral infarction (lacunar) (P> 0.05) -1 genotypes, KK genotype in cerebral infarction group was significantly more than that in control group, EE genotype was slightly lower than that in control group, the difference was statistically significant (P <0.05); There was no significant difference in EK genotype between the two groups (P> 0.05). Compared with the control group, the allele distribution in the two groups was significantly higher than that in the control group (P <0.05). Conclusion There is a clear correlation between polymorphism of ICAM-1K469E gene and ICAM-1 in patients with cerebral infarction in Han nationality. E allele may be an important genetic factor affecting cerebral infarction in Han nationality.
