目的:研究不同剂量DF-521(东菱克栓酶,Batroxobin)、低分子量肝素(LMWH)对体外培养人正常大隐静脉片内膜增生的影响。方法:体外培养人正常大隐静脉片,加入不同剂量DF-521(10-4BU/ml,10-2BU/ml)和低分子量肝素(0.108IU/ml,10.8IU/ml)。培养最后4d掺入5-溴脱氧尿核苷(5-BrdU)。应用免疫组织化学方法行抗5-BrdU单克隆抗体ABC法染色及抗人平滑肌肌动蛋白(α-Actin)单克隆抗体与Verhoef弹力纤维联合染色。并应用计算机图像分析仪进行图像分析,显微镜细胞计数等方法观察内膜增生及细胞增殖。结果:培养静脉片可见明显内膜增生和平滑肌细胞(SMC)增殖。与对照组相比,较高浓度DF-521及低分子量肝素明显抑制培养血管片内膜增生。对照组内膜厚度(27±3.40)μm,LMWH(7±1.20)μm,DF-521(6±1.23)μm,P<0.001。对照组平滑肌细胞增殖指数(30±2.20)%,LMWH(6±1.20)%,DF-521(5±1.74)%,P<0.001。两种药物作用效果相近。结论:DF-521及低分子量肝素可抑制移植血管内膜增生及平滑肌?
Objective: To study the effects of different doses of DF-521 (Batroxobin) and LMWH on intima hyperplasia of human normal saphenous vein in vitro. Methods: Normal human saphenous vein was cultured in vitro. Different doses of DF-521 (10-4BU / ml, 10-2BU / ml) and low molecular weight heparin (0.108IU / ml, 10.8IU / ml) were added. The last 4 days of culture incorporates 5-bromodeoxyuridine (5-BrdU). Immunohistochemical staining was performed by ABC staining of anti-5-BrdU monoclonal antibody and anti-α-actin monoclonal antibody and Verhoef elastic fibers. The application of computer image analyzer for image analysis, microscopic cell counting and other methods to observe intimal hyperplasia and cell proliferation. Results: Intimal hyperplasia and smooth muscle cell (SMC) proliferation were observed in cultured venous. Compared with the control group, higher concentrations of DF-521 and low molecular weight heparin significantly inhibited intimal hyperplasia of cultured blood vessels. The thickness of the control group was 27 ± 3.40 μm, LMWH was 7 ± 1.20 μm, DF-521 was 6 ± 1.23 μm, P <0.001. The proliferation index of smooth muscle cells in control group (30 ± 2.20)%, LMWH (6 ± 1.20)% and DF-521 (5 ± 1.74)%, P <0.001. The effect of the two drugs are similar. Conclusion: DF-521 and low molecular weight heparin can inhibit vascular intimal hyperplasia and smooth muscle graft?
