目的揭示巨噬细胞集落刺激因子(M-CSF)的抗动脉粥样硬化作用是否与其保护单核细胞免受过氧化损伤有关。材料与方法用富含M-CSF的L929细胞条件培养液(L929-CM)作为M-CSF来源,以凝胶电泳、DNA断裂定量等方法观察了M-CSF对叔丁基氢过氧化物(tbOOH)诱导的U937细胞DNA断裂损伤的影响。结果tbOOH可以引起U937细胞的DNA断裂损伤,且损伤随tbOOH作用时间的延长而趋明显;而L929-CM可以减轻tbOOH诱发的U937细胞DNA断裂损伤。结论M-CSF对单核细胞具有一定的保护作用,可减轻其氧化损伤。
Objective To investigate whether the anti-atherosclerotic effect of macrophage colony-stimulating factor (M-CSF) is related to its ability to protect monocytes from peroxidative damage. MATERIALS AND METHODS M-CSF-derived L929-CM was used as M-CSF source. M-CSF t-butyl hydroperoxide (tbOOH) was observed by gel electrophoresis and DNA fragmentation. Induced DNA damage in U937 cells. Results tbOOH could induce the DNA damage in U937 cells, and the damage became obvious with the prolongation of tbOOH. However, L929-CM could reduce the DNA damage of U937 cells induced by tbOOH. Conclusion M-CSF can protect monocytes from oxidative damage.
