目的:基于NF-κB信号通路观察小青龙汤合玉屏风散对变应性鼻炎大鼠鼻黏膜AQP5含量和AQP5 m RNA表达的影响,揭示小青龙汤合玉屏风散治疗AR的作用机制。方法:将清洁级健康雄性Wistar大鼠48只按体重采用随机区组法分为6组,即分成正常组、AR模型组、Forskolin(c AMP激活剂)干预组、H89(PKA抑制剂)干预组、PDTC(NF-κB抑制剂)干预组、小青龙汤合玉屏风散组,每组8只。采用卵蛋白全身致敏与局部攻击方法制作AR大鼠模型,观察小青龙汤合玉屏风散组治疗后鼻黏膜AQP5含量和AQP5 m RNA表达,结果进行统计学分析。结果:模型组AQP5含量和AQP5 m RNA表达下调,c AMP激活剂Forskolin干预组、NF-κB抑制剂PDTC干预组、小青龙汤合玉屏风散治疗组均能使AQP5含量和AQP5 m RNA表达上调,而PKA抑制剂H89能下调AQP5含量和AQP5 m RNA表达。结论:小青龙汤合玉屏风散治疗变应性鼻炎作用机制可能与抑制NF-κB信号通路和兴奋c AMP-PKA信号通路使AQP5含量和AQP5 m RNA表达上调有关。
OBJECTIVE: To observe the effect of Xiaolonglong Decoction and Yupingfeng Powder on the expression of AQP5 mRNA and the expression of AQP5 mRNA in nasal mucosa of allergic rhinitis rats based on the NF-κB signaling pathway, and to reveal the mechanism of Xiaolonglong Decoction combined with Yupingfeng Powder in treating AR. Methods: Forty-eight healthy male Wistar rats were divided into 6 groups according to body weight: normal group, AR model group, Forskolin intervention group, H89 (PKA inhibitor) intervention group Group, PDTC (NF-κB inhibitor) intervention group, Xiaolonglong Decoction combined with Yupingfengsan group, 8 rats in each group. A rat model of AR was established by systemic sensitization and local challenge with ovalbumin. AQP5 expression and AQP5 mRNA expression in nasal mucosa were observed after treatment with Xiaolonglong Decoction and Yupingfengsan. The results were statistically analyzed. Results: The levels of AQP5 and AQP5 mRNA in model group were down-regulated. The expressions of AQP5 and AQP5 mRNA were up-regulated in Forskolin intervention group, NF-κB inhibitor PDTC intervention group and Xiaoqinglong decoction combined with Yupingfengsan treatment group PKA inhibitor H89 downregulated AQP5 expression and AQP5 mRNA expression. CONCLUSION: The mechanism of Xiao Qing Long Decoction combined with Yupingfeng Powder in treating allergic rhinitis may be related to the upregulation of AQP5 mRNA and the expression of AQP5 and the inhibition of NF-κB signal pathway and excited AMPA-PKA signaling pathway.
