目的:探讨升清降糖合剂(简称SQ)对糖尿病大鼠空腹血糖水平和胰岛细胞形态及功能的影响。方法:腹腔注射链脲佐菌素(STZ)诱导建立糖尿病大鼠模型。根据不同的干预方式将糖尿病大鼠分为模型对照组、SQ低剂量组、SQ中剂量组、SQ高剂量组。每两周测空腹血糖和体重,于干预后第八周末,各组大鼠麻醉后采集血样,ELISA法测定血清C肽水平,取胰腺组织HE染色观察病理学改变。结果:空腹血糖水平,模型对照组(26.92±3.68)>SQ低剂量组(23.87±2.86)>SQ高剂量组(20.66±2.81)>SQ中剂量组(20.66±2.81)>正常对照组(4.55±0.57),SQ中、高剂量与模型对照组比较差异有统计学意义(P<0.05)。血清C肽水平,正常对照组(2.27±0.47)>SQ中剂量组(1.76±0.91)>SQ高剂量组(1.74±0.79)>SQ低剂量组(1.11±0.36)>模型对照组(0.70±0.62),SQ中、高剂量组与模型对照组比较差异有统计学意义(P<0.05)。胰岛病理切片形态学观察发现,模型对照组大鼠胰岛细胞数量减少,排列紊乱,胞质呈空泡状,部分细胞核固缩;SQ各组大鼠胰岛细胞受损程度较轻。结论 SQ能明显降低糖尿病大鼠的空腹血糖水平,提高血清C肽水平,保护胰岛细胞功能。
Objective: To investigate the effects of Shengqing Jiangtang Mixture (SQ) on fasting blood glucose and islet cell morphology and function in diabetic rats. Methods: Diabetic rats were induced by intraperitoneal injection of streptozotocin (STZ). According to different interventions, diabetic rats were divided into model control group, SQ low dose group, SQ middle dose group and SQ high dose group. Fasting blood glucose and body weight were measured every two weeks. At the end of the eighth week after the intervention, blood samples were collected from rats in each group. Serum C-peptide levels were measured by ELISA. Pathological changes were observed by HE staining. Results: The level of fasting blood glucose in the model control group (26.92 ± 3.68)> SQ low dose group (23.87 ± 2.86)> SQ high dose group (20.66 ± 2.81)> SQ medium dose group (20.66 ± 2.81)> normal control group (4.55 ± 0.57). There was significant difference between the medium and high doses of SQ and the model control group (P <0.05). Serum C-peptide levels in the normal control group (2.27 ± 0.47)> SQ medium dose group (1.76 ± 0.91)> SQ high dose group (1.74 ± 0.79)> SQ low dose group (1.11 ± 0.36)> model control group 0.62). There was significant difference between SQ and high dose group and model control group (P <0.05). The morphological observation of islet pathological sections showed that the number of islet cells in the model control group was reduced and disordered, the cytoplasm was vacuolated, and some nuclei were pyknotic. The damage of pancreatic islets in SQ group was less. Conclusion SQ can significantly reduce fasting blood glucose level, increase serum C-peptide level and protect islet cell function in diabetic rats.
