目的:探讨lncRNA AC007009.1在非小细胞肺癌(non-small cell lung cancer,NSCLC)组织中的表达及其临床意义。方法:收集2011年6月至2016年6月四川省自贡市第四人民医院105例NSCLC患者;通过实时荧光定量PCR法检测lncRNA AC007009.1在105例NSCLC组织、72例癌旁组织及33正常肺组织中的表达水平,分析lncRNA AC007009.1的表达与患者部分临床病理学特征之间的关系,了解lncRNA AC007009.1的表达与患者OS和PFS关系及对患者预后的影响。结果:LncRNA AC007009.1在105例NSCLC癌组织中的表达水平明显高于癌旁组织及正常肺组织(5.30±1.96 vs 1.01±0.33、0.99±0.29,均P<0.01),LncRNA AC007009.1的表达水平与患者癌症分期、远处转移、病理类型及生存状态明显有关(P<0.05)。高表达lncRNA AC007009.1患者的OS及PFS较低表达患者明显缩短(P<0.01),Cox多因素回归模型分析显示,lncRNA AC007009.1的表达水平、淋巴远处转移及临床分期是NSCLC独立的预后影响因素(P<0.05)。结论:lncRNA AC007009.1参与调节NSCLC的发生发展,可作为潜在的NSCLC诊断和预后评估的分子标志物。
Objective: To investigate the expression of lncRNA AC007009.1 in non-small cell lung cancer (NSCLC) and its clinical significance. Methods: From June 2011 to June 2016, 105 cases of NSCLC were collected from the Fourth People’s Hospital of Zigong City, Sichuan Province. Real-time fluorescence quantitative PCR was used to detect the expression of lncRNA AC007009.1 in 105 NSCLC tissues, 72 paracancerous tissues and 33 normal Lung tissue, analyze the relationship between the expression of lncRNA AC007009.1 and some clinicopathological features of patients to understand the relationship between the expression of lncRNA AC007009.1 and OS and PFS and the prognosis of patients. Results: The expression level of LncRNA AC007009.1 in 105 NSCLC tissues was significantly higher than that in adjacent non-cancerous tissues and normal lung tissues (5.30 ± 1.96 vs 1.01 ± 0.33, 0.99 ± 0.29, all P <0.01), and LncRNA AC007009.1 The expression level was significantly correlated with cancer stage, distant metastasis, pathological type and survival status (P <0.05). The expression of lncRNA AC007009.1 in patients with high expression of lncRNA AC007009.1 was significantly shortened (P <0.01), and the expression of lncRNA AC007009.1, lymph node metastasis and clinical stage were independent of NSCLC Prognostic factors (P <0.05). CONCLUSION: lncRNA AC007009.1 is involved in the regulation of the development of NSCLC and may be used as a potential molecular marker for the diagnosis and prognosis of NSCLC.
