目的 :研究抗氧化剂与钙离子拮抗剂联合应用对大鼠免疫性睾丸炎的防治作用及其作用机制。 方法 :Wistar大鼠随机分为正常对照组、预防免疫性睾丸炎症组 (预防组 )和免疫性睾丸炎症模型组 (模型组 ) ,从第一次免疫的次日起 ,预防组每天给予 90 m g/kg维生素 E、C合剂 (Vit C∶ Vit E=2∶ 1)和 5 0 m g/kg维拉帕米灌胃 ,共80 d,模型组未给予干预措施 ,正常对照组未加任何处理。检测 3组的精子计数、精子畸形率、睾丸重量、曲细精管直径 ,光镜观察睾丸间质及曲细精管生精细胞的变化。 结果 :预防组与正常对照组的精子计数、精子畸形率、睾丸重量和曲细精管直径差异均无统计学意义 (P >0 .0 5 ) ,镜下表现接近于正常对照组。模型组与正常对照组相比 ,精子计数、睾丸重量和曲细精管内径均有明显减少 (P <0 .0 5~ 0 .0 1) ,精子畸形率明显增加 (P <0 .0 1)。光镜下睾丸间质明显充血 ,生精细胞脱落 ,生精上皮空泡变 ,多核巨精细胞形成 ,不同程度的炎性细胞浸润。 结论 :维生素 E、维生素 C、维拉帕米联合应用可明显减轻大鼠免疫性炎症损伤。
Objective: To study the preventive and therapeutic effects of antioxidants and calcium antagonists on immune orchitis in rats and its mechanism. Methods: Wistar rats were randomly divided into normal control group, prophylactic immunization testicular inflammation group (prophylaxis group) and immune testicular inflammation model group (model group). From the day after the first immunization, prophylaxis group received daily 90 mg / kg vitamin E, C mixture (Vit C: Vit E = 2: 1) and verapamil at 50 mg / kg for 80 days. No intervention was given in the model group and no treatment was taken in the normal control group. The sperm count, sperm deformity rate, testicular weight and seminiferous tubule diameter were measured in 3 groups. The changes of spermatogenic cells in testis interstitium and seminiferous tubules were observed with light microscope. Results: There was no significant difference in sperm count, sperm deformity, testicular weight and seminiferous tubule diameter between the prophylaxis group and the normal control group (P> 0.05). The microscopic findings were similar to those of the normal control group. Compared with the normal control group, the sperm count, testicular weight, and seminiferous tubule diameter were significantly decreased in the model group (P <0.05-0.01) and the sperm deformity rate was significantly increased (P <0.01) ). Under light microscope, there was obvious hyperemia of testicular stroma, spermatogenic cells shedding, vacuoles of germinal epithelium, formation of multinucleated giant cells and infiltration of inflammatory cells to varying degrees. Conclusion: The combination of vitamin E, vitamin C and verapamil can obviously reduce the inflammatory injury in rats.
