目的:探究阿米替林和吗啡联合应用对尼古丁依赖-戒断小鼠的镇痛效果。方法昆明小鼠72只,分为生理盐水组(n=12)和尼古丁依赖-戒断组(n=60),尼古丁依赖-戒断组小鼠皮下注射尼古丁7天制备尼古丁依赖-戒断模型。成模小鼠采用随机数字表法随机分为模型组、吗啡组、低阿米组(吗啡+20 mg/kg阿米替林)、中阿米组(吗啡+30 mg/kg阿米替林)以及高阿米组(吗啡+40 mg/kg阿米替林)5组,每组12只。给药后采用热板法观察各组小鼠舔足反应潜伏期的变化。结果与模型组比较:各给药组小鼠舔足反应潜伏期均明显延长(P<0.01);与吗啡组比较,中阿米组和高阿米组小鼠给药后5、15、45 min的舔足反应潜伏期均延长(P<0.05),而低阿米组小鼠仅在给药后15 min的舔足反应潜伏期延长(P<0.05);与低阿米组比较,高阿米组小鼠给药后5、15、45 min的舔足反应潜伏期均延长(P<0.05),而中阿米组小鼠只有在给药后45 min的舔足反应潜伏期延长(P<0.05);与中阿米组比较,高阿米组小鼠给药后5 min和45 min的舔足反应潜伏期均延长(P<0.05),而给药后15 min虽然舔足反应潜伏期延长,但差异没有统计学意义( P>0.05)。结论一定剂量的阿米替林与吗啡联合使用较单独使用吗啡的镇痛效果好,加大阿米替林的剂量,小鼠的舔足反应潜伏期相应延长。“,”Objective To investigate the analgesic effects of amitriptyline combined with morphine on nicotine de-pendent mice with withdrawal syndrome.Methods A total of 72 mice were divided into a normal saline group ( n=12) and a nicotine dependence-withdrawal group ( n=60 ) .The mice in the nicotine dependence-withdrawal group were subcutaneously injected with nicotine for seven days.The resultant modeled mice were randomly divided into a model group, a morphine group, a low-dose amitriptylin group ( morphine +20 mg/kg amitriptyline) , a medium-dose ami-triptylin group ( morphine +30 mg/kg amitriptyline) and a high-dose amitriptylin group ( morphine +40 mg/kg ami-triptyline) (n=12).After administration, the hot-plate test was used to observe the changes in the response latency of licking behavior.Results Compared with the model group, the latency of licking behavior was significantly prolonged in each treatment group (P<0.01).Compared with the morphine group, the low-dose amitriptylin group presented ex-tended latency 15 min after administration (P<0.05), but the medium-and high-dose amitriptylin groups showed ex-tended latency 5, 15 and 45 min after administration (P<0.05).Compared with the low-dose amitriptylin group, the medium-dose amitriptylin group presented extended latency 45 min after administration (P<0.05), but the high-dose amitriptylin group showed extended latency 5, 15 and 45 min after administration (P<0.05).Compared with the medi-um-dose amitriptylin group, the high-dose amitriptylin group presented extended latency 5 and 45 min after adminis-tration (P<0.05).Conclusion A certain dose of amitriptyline combined with morphine can produce better analgesiceffects than morphine alone.An increased dose of amitriptyline can result in prolonged latency of licking response in mice.
