目的鉴定长沙市辖区内2012年-2015年分离的布鲁菌种型,掌握长沙市布鲁菌病感染病原体种型分布以及长沙市的布病流行趋势,为今后布鲁菌病防控策略提供病原学参考。方法采用传统的病原菌检测方法对37株分离自长沙市区哨点医院的患者血液标本的可疑布鲁菌株进行分离培养鉴定后,纯化培养细菌并提取核酸DNA作为模板,采用AMSO聚合酶链法布鲁菌种型特异性PCR(AMSO-PCR)对这些布鲁菌株进行分子种型鉴定。结果经传统病原菌检测方法分离培养鉴定,37株菌株均为布鲁菌。AMSO-PCR分子鉴定结果表明,37株布鲁菌均为羊种布鲁菌,未见其他型别。结论 2012年-2015年长沙市人间流行的布鲁菌均为羊种布鲁菌。
Objective To identify the types of brucellosis isolated in Changsha area from 2012 to 2015 and to grasp the distribution of brucellosis-infected pathogens in Changsha City and the epidemic trend of brucellosis in Changsha City, so as to provide the prevention and control strategies for brucellosis in the future Etiological reference. Methods The traditional method of pathogen detection was used to isolate and identify 37 suspected strains of Brucella isolated from the blood samples of sentinel hospitals in Changsha City. The bacteria were purified and DNA was extracted and used as templates. AMSO PCR The Brucella species-specific PCR (AMSO-PCR) was used to characterize these Brucella strains. Results Isolation and identification by traditional pathogen detection method, 37 strains are Brucella. AMSO-PCR molecular identification results show that all of the 37 strains of Brucella are Brucella brucei, no other type. CONCLUSIONS: Brucella spp. Circulating in Changsha City from 2012 to 2015 are all Brucella abortus.
