目的:通过大鼠肝微粒体体外代谢系统考察氧化苦参碱对CYP2C19酶活性的影响。方法:采用本实验室建立的大鼠肝微粒体体外代谢系统,用RP-HPLC法测定CYP2C19酶活性,通过考察加入和不加入氧化苦参碱时CYP2C19酶活性的变化,分析氧化苦参碱对CYP2C19酶活性的影响。结果:加入氧化苦参碱后CYP2C19酶活性较不加入氧化苦参碱前降低了(35.39±5.66)%(P<0.01),差别有统计学意义。结论:氧化苦参碱对CYP2C19酶活性有显著抑制作用,故临床联合用药时应注意代谢性相互作用对疗效的影响。
OBJECTIVE: To investigate the effect of oxymatrine on the activity of CYP2C19 in rat liver microsomes in vitro. Methods: The rat liver microsome metabolic system established in our laboratory was used to measure the activity of CYP2C19 by RP-HPLC. The changes of CYP2C19 activity were analyzed by the changes of CYP2C19 activity with and without addition of oxymatrine CYP2C19 enzyme activity. Results: After adding oxymatrine, the activity of CYP2C19 decreased (35.39 ± 5.66)% (P <0.01) compared with that before adding oxymatrine, the difference was statistically significant. Conclusion: Oxymatrine has a significant inhibitory effect on the activity of CYP2C19, so the effect of metabolic interaction on the curative effect should be noticed in clinical combination therapy.
