目的探讨麻桂药浴方熏蒸对腺嘌呤致CRF大鼠模型的干预效果,及对MMPs活力的影响。方法将96只SD大鼠随机分为A、B、C、D、E、F组,每组16只。A、B、C、D、E组均予以腺嘌呤致CRF模型,A、B、C组给予麻桂药浴方水煎液40、20、10 ml熏蒸,D组自来水40 ml熏蒸,E、F组不熏蒸,均干预12周。比较干预后血清BUN、Cr、HCT、PLT、GP-ⅡbⅢa水平,行肾脏组织学检查及肾小管评分,行肾组织MMP-2、MMP-9、TIMP-1和TIMP-2 mRNA及蛋白,LN、Ⅳ-C蛋白检测。结果 D、E组血清Cr、BUN及GP-ⅡbⅢa水平,肾小管评分,TIMP-1、TIMP-2蛋白,LN、Ⅳ-C蛋白均高于A、B、C、F组,HCT、PLT水平,MMP-2、MMP-9 mRNA及蛋白均低于A、B、C、F组(P<0.05或P<0.01)。结论麻桂药浴方熏蒸对腺嘌呤致CRF大鼠模型具有较好的保护作用,可能与增加MMPs活力有关。
Objective To investigate the intervention effect of Ma Gui medicinal bath fumigation on adenine-induced CRF rat model and its effect on the activity of MMPs. Methods 96 Sprague-Dawley rats were randomly divided into A, B, C, D, E and F groups, with 16 rats in each group. A, B, C, D, E group were given adenine caused by CRF model, A, B, C group was given Ma Guiyao bath decoction 40,20,10 ml fumigation, D group tap water 40 ml fumigation, Group F did not fumigate, both interventions for 12 weeks. The levels of serum BUN, Cr, HCT, PLT and GP-IIbIIIa in renal tissues were measured. The expressions of MMP-2, MMP-9, TIMP-1 and TIMP- , Ⅳ-C protein detection. Results The serum levels of Cr, BUN and GP-ⅡbⅢa, tubule score, TIMP-1, TIMP-2, LN and Ⅳ-C in groups D and E were higher than those in groups A, B, C and F , MMP-2 and MMP-9 mRNA and protein were lower than those in A, B, C and F groups (P <0.05 or P <0.01). Conclusion Flue-cured Cucumber Decoction has better protective effect on adenosine-induced CRF rat model, which may be related to increasing the vitality of MMPs.
