目的:观察腺苷预处理对心肌缺血再灌注损伤的影响,探讨血红素加氧酶-1(HO-1)在其中的作用。方法:雄性SD大鼠18只,体重(250±16)g,随机分为3组,每组6只:①对照组(C组),②缺血再灌注组(IR组),③腺苷预处理+缺血再灌注组(AP组)。采用结扎/放松左冠状动脉前降支建立心肌缺血再灌注损伤模型。再灌注结束后,采用免疫组化法检测心肌HO-1变化,检测心肌梗死面积、心肌组织丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性,及血清TNF-α水平。结果:与C组比较,IR组心肌组织MDA含量和血清TNF-α水平升高,SOD活性降低,心肌梗死面积增加(P<0.05),HO-1表达差异无统计学意义(P>0.05);与IR组比较,AP组心肌组织MDA含量和血清TNF-α水平降低,SOD活性增强,心肌梗死面积减少,HO-1表达增加(P<0.05)。结论:腺苷预处理对心肌缺血再灌注损伤的保护作用与HO-1的表达增加有关。
Objective: To observe the effect of adenosine preconditioning on myocardial ischemia-reperfusion injury and to explore the role of heme oxygenase-1 (HO-1) in it. Methods: Totally 18 male Sprague-Dawley rats weighing 250 ± 16 g were randomly divided into 3 groups with 6 rats in each group: ① control group (C), ② ischemia-reperfusion group (IR), ③ adenosine Pretreatment + ischemia-reperfusion group (AP group). A myocardial ischemia-reperfusion injury model was established by ligating / relaxing the left anterior descending coronary artery. After reperfusion, the changes of HO-1 in myocardium were detected by immunohistochemistry. The area of myocardial infarction, the content of malondialdehyde (MDA), the activity of superoxide dismutase (SOD) and the level of serum TNF-α in myocardial tissue were detected. Results: Compared with group C, the content of MDA and the level of serum TNF-α, the activity of SOD and the area of infarcted myocardium in IR group were increased (P <0.05), while there was no significant difference in HO-1 expression (P> 0.05) Compared with IR group, the content of MDA and the level of serum TNF-α in myocardium of AP group were decreased, the activity of SOD was increased, the area of myocardial infarction was decreased and the expression of HO-1 was increased (P <0.05). Conclusion: The protective effect of adenosine preconditioning on myocardial ischemia-reperfusion injury is related to the increase of HO-1 expression.
