目的利用常见呼吸道感染细菌制备的联合疫苗作用人外周血单个核细胞,探讨其对机体免疫功能的影响,为临床应用提供参考。方法分离培养引起我国呼吸道感染的最常见细菌,利用超声波破碎,硫酸铵沉淀蛋白,构建成细菌疫苗,体外作用人外周血单个核细胞,采用CCK-8法测定其增殖活性;酶联免疫吸附试验(ELISA)测细胞因子白细胞介素-2(IL-2)和干扰素-γ(IFN-γ)含量;流式细胞仪(FCM)检测细胞表型变化。结果联合疫苗组单个核细胞增殖活性显著高于空白对照组(P<0.01);联合疫苗组IL-2、IFN-γ的浓度比空白对照组高,并随疫苗浓度的增加而增多,在一定浓度范围内成剂量依赖关系;细胞表型分析显示联合疫苗既能刺激T细胞增殖,也能刺激B细胞增殖。结论此联合疫苗可增强免疫功能,它能提高细胞免疫功能,对体液免疫功能也具有明显影响。
OBJECTIVE: To study the effects of combined vaccine prepared by common respiratory tract infection on peripheral blood mononuclear cells (PBMCs) on the immune function and provide a reference for clinical application. Methods The most common bacteria that cause respiratory tract infection in China were isolated and cultured. The cells were treated with ultrasonic wave and precipitated with ammonium sulfate to establish a bacterial vaccine. Human peripheral blood mononuclear cells (PBMCs) were treated with CCK-8 in vitro. Enzyme-linked immunosorbent assay (IL-2) and interferon-γ (IFN-γ) were measured by enzyme linked immunosorbent assay (ELISA). Cell phenotypes were detected by flow cytometry (FCM) Results The proliferation activity of mononuclear cells in the combination vaccine group was significantly higher than that in the blank control group (P <0.01). The concentrations of IL-2 and IFN-γ in the combination vaccine group were higher than those in the blank control group and increased with the increase of the vaccine concentration. Dose-dependent concentration range; cell phenotype analysis showed that the combination vaccine can stimulate T cell proliferation, but also stimulate B cell proliferation. Conclusion The combination vaccine can enhance the immune function, which can improve the cellular immune function and have a significant effect on humoral immune function.
