目的 观察2 甲氧基雌二醇对骨髓瘤细胞系CZ 1细胞诱导分化作用。方法 通过细胞形态观察,细胞表面标志分析和细胞分泌轻链蛋白水平的测定,观察2 甲氧基雌二醇对CZ 1细胞的诱导分化作用。结果 CZ 1细胞经0. 1~0. 5μmol/L2 甲氧基雌二醇作用48h后细胞形态向成熟阶段发展,表现为细胞胞核缩小,胞浆丰富,核浆比例下降,核仁减少或消失,核染色质变粗、变密。细胞表面标志CD49e阳性表达率由(12. 20±1. 57)%增加到( 24. 80±1. 26 )%,差异有统计学意义(P<0. 05); CZ 1细胞分泌轻链蛋白由(35. 97±2. 60)μg/ml升高到(79. 67±1. 88)μg/ml,差异有统计学意义(P<0. 05)。结论 较低浓度2 甲氧基雌二醇可诱导骨髓瘤细胞向成熟阶段分化。
Objective To observe the effect of 2 methoxyestradiol on differentiation of myeloma cell line CZ 1. Methods Morphological observation, cell surface marker analysis and determination of cell-secreted light chain protein levels were used to observe the differentiation of CZ 1 cells induced by 2-methoxyestradiol. Results CZ 1 cells were exposed to 0.1-1.5 μmol / L methoxyestradiol for 48 h, and then the morphology of CZ 1 cells progressed to mature stage. The results showed that the cell nucleus was reduced, the cytoplasm was rich, the ratio of nucleus pulposus decreased, the number of nucleolus decreased or Disappeared, nuclear chromatin thicker, dense. The positive rate of CD49e expression on the cell surface was increased from (12.20 ± 1.57)% to (24.88 ± 1.26)%, the difference was statistically significant (P <0.05); CZ 1 cells secreted the light chain The protein level increased from (35.97 ± 2.60) μg / ml to (79.67 ± 1.88) μg / ml, the difference was statistically significant (P <0.05). Conclusions Lower concentration of 2-methoxyestradiol can induce myeloma cells to differentiate into mature stage.
