目的:研究钩藤总碱对大鼠肝脏和肝功能的影响,为探讨钩藤总碱肝毒性提供实验依据。方法:选用健康7周龄Wistar大鼠40只,雌雄各半,随机分为对照组、钩藤总碱小剂量组、中剂量组和大剂量组,分别给予生理盐水、2.5mg/kg、10mg/kg和15mg/kg钩藤总碱水溶液,于给药后第12周处死取颈动脉血,检测肝脏各项指标。结果:各组大鼠肝指数差异比较有统计学意义(P<0.05),钩藤总碱各剂量组肝指数均高于对照组,其中钩藤总碱大剂量组肝指数最高;各组大鼠血脂指标TG、LDL-C和HDL-C差异比较无统计学意义(P>0.05);钩藤总碱各剂量组ALT和AST水平均高于对照组,其中钩藤总碱大剂量组ALT和AST水平最高,而各组TP、ALB和T-BIL差异比较无统计学意义(P>0.05)。结论:钩藤总碱具有肝毒性作用,故在临床治疗时应合理用药。
OBJECTIVE: To study the effects of rhynchophyllines on the liver and liver function in rats, and to provide experimental basis for exploring the hepatotoxicity of rhynchophylla total alkaloids. Methods: Forty healthy Wistar rats aged 7 weeks were randomly divided into control group, low dose group of Uncaria total, middle dose group and high dose group, and were given normal saline, 2.5mg / kg, 10mg / kg and 15mg / kg Uncaria total alkaloids aqueous solution were sacrificed at the 12th week after administration to take the carotid artery, liver indicators were measured. Results: The difference of liver index in each group was statistically significant (P <0.05), and the liver index of each group was higher than that of the control group There was no significant difference in TG, LDL-C and HDL-C between the two groups (P> 0.05). The levels of ALT and AST in each group were higher than those in the control group And AST levels were the highest, while the differences of TP, ALB and T-BIL in each group were not statistically significant (P> 0.05). Conclusion: Uncaria total alkaloids have hepatotoxicity, so it should be used properly in clinical treatment.
