[目的]探讨蛋白酶体抑制剂硼替佐米(bortezomib)对K562细胞株DNA甲基转移酶(DNMTs)表达、细胞凋亡的影响。[方法]以不同浓度的硼替佐米(0、6、20、60nmol/L)处理白血病K562细胞株,Western blot法检测细胞内DNMT1的表达,流式细胞术检测细胞凋亡。[结果]与对照组相比,硼替佐米可显著抑制DNMT1表达;硼替佐米作用于细胞12、24、36h后细胞凋亡率逐渐增加,60nmol/L硼替佐米作用36h后细胞凋亡率最高,为61.68%±3.20%。[结论]硼替佐米抑制K562细胞株DNMT1表达,并诱导细胞凋亡,呈浓度依赖性。
[Objective] To explore the effect of proteasome inhibitor bortezomib on DNA methyltransferase (DNMTs) expression and apoptosis in K562 cell line. [Method] Leukemia K562 cells were treated with different concentrations of bortezomib (0, 6, 20 and 60 nmol / L). The expression of DNMT1 in cells was detected by Western blot and the apoptosis was detected by flow cytometry. [Results] Compared with the control group, bortezomib could significantly inhibit the expression of DNMT1. The apoptosis rate of the cells treated with bortezomib increased gradually at 12, 24 and 36 hours. The apoptosis rate of the cells treated with 60 nmol / L bortezomib for 36 hours The highest was 61.68% ± 3.20%. [Conclusion] Bortezomib inhibits DNMT1 expression and induces apoptosis in K562 cells in a concentration-dependent manner.
