目的:了解新生小鼠耳蜗在体外条件下存活状况,为体外急性实验提供组织模型。方法:对出生1~3d小鼠耳蜗以体外悬浮培养方法,并用扫描电镜对其外毛细胞的结构和形态进行观察。结果:采用这种方法培养的外毛细胞,在存活的20h内,其细胞形态和结构无明显改变。结论:耳蜗悬浮培养对短期离体外毛细胞的研究不失为稳定可靠的方法,而扫描电镜也补充证实为一种有效、简便的观察手段
Objective: To understand the survival of neonatal mouse cochlea in vitro and to provide a tissue model for acute in vitro experiments. Methods: The cochleas of 1 ~ 3 days old mice were cultured in vitro and the structure and morphology of their outer hair cells were observed by scanning electron microscopy. Results: The outer hair cells cultured by this method showed no significant changes in cell morphology and structure within 20 hours of survival. Conclusion: Cochlear suspension culture of short hair cells in vitro after the study is a stable and reliable method, and scanning electron microscopy also confirmed as an effective and easy means of observation
