目的观察microRNA-122(miR-122)在利福平诱导的小鼠肝损伤中的动态变化并探讨其与抗结核药物性肝损伤(ADLI)的关系。方法将48只昆明小鼠随机分为对照组(8只)和实验组(40只)。实验组每天予以利福平(135mg/kg)灌胃处理,对照组每天给予同等容积生理盐水灌胃,其中实验组分别在给药后的1、3、5、7、14d后采集8只小鼠的肝组织和血清标本。采用全自动生化分析仪检测血清谷丙转氨酶(ALT)和谷草转氨酶(AST)的表达水平,实时荧光定量PCR分析miR-122的表达。结果随着给药时间延长,miR-122整体表达呈下降趋势,给药3d后发生显著性变化(P<0.05)。ALT和AST在给药后呈上升趋势,均在14d后出现显著性改变(P<0.05)。相关性分析结果显示miR-122与ALT和肝组织病理学评分呈负相关(r=-0.365,r=-0.609,均P<0.01)。结论 miR-122与利福平所致的肝损伤的发生存在相关性。
Objective To investigate the dynamic changes of microRNA-122 (miR-122) in rifampin-induced liver injury in mice and its relationship with anti-tuberculosis drug-induced liver injury (ADLI). Methods 48 Kunming mice were randomly divided into control group (n = 8) and experimental group (n = 40). Rifampicin (135mg / kg) was given to the experimental group intragastrically every day, while the control group was given the same volume of normal saline intragastrically every day, in which the experimental group were collected after the 1,3,5,7,14 d Rat liver and serum samples. The levels of serum ALT and AST were detected by automatic biochemical analyzer. The expression of miR-122 was analyzed by real-time fluorescence quantitative PCR. Results With the prolongation of administration time, the overall expression of miR-122 showed a decreasing trend, which was significantly changed after 3d (P <0.05). ALT and AST showed an upward trend after administration, both of which showed significant changes after 14 days (P <0.05). Correlation analysis showed that miR-122 was negatively correlated with ALT and liver histopathological score (r = -0.365, r = -0.609, all P <0.01). Conclusion There is a correlation between the expression of miR-122 and rifampicin-induced liver injury.
