目的:探讨反义核酸和维甲酸对早幼粒白血病细胞株NB_4细胞生长分化的影响.方法:采用细胞生长曲线、形态学、细胞表面标志及硝基四氮唑蓝(NBT)还原试验判定NB_4细胞的生长、分化及功能.结果:PML-RARα基因的反义核酸和全反式维甲酸能够诱导NB_4细胞分化,提高NBT还原率,降低细胞表面的CD33抗原,升高CD11b抗原.反义核酸更有利于CD33抗原的降低,而全反式维甲酸更有利于CD11b抗原的升高.结论:PML-RARα基因的反义核酸和全反式维甲酸能诱导NB_4细胞分化,但它们的作用机制可能不同.
OBJECTIVE: To investigate the effect of antisense nucleic acids and retinoids on the growth and differentiation of promyelocytic leukemia cell line NB_4. METHODS: Cell growth curves, morphology, cell surface markers, and nitroblue tetrazolium (NBT) reduction test were used to determine NB_4 Cellular growth, differentiation and function. Results: Antisense nucleic acids of PML-RARα and all-trans retinoic acid can induce differentiation of NB_4 cells, increase NBT reduction rate, reduce cell surface CD33 antigen, and increase CD11b antigen. It is more conducive to the reduction of CD33 antigen, and all-trans retinoic acid is more conducive to the elevation of CD11b antigen. Conclusion: The antisense nucleic acid of PML-RARα gene and all-trans retinoic acid can induce differentiation of NB_4 cells, but their mechanism of action May be different.
