目的:研究壮骨伸筋胶囊(Zhuanggu Shenjin capsules,ZSJ)对去卵巢骨质疏松大鼠骨密度及骨代谢的影响,并探讨其相关的作用机制。方法:将72只SD大鼠按随机分为假手术组(12只)和造模组(60只),模型组采用摘除大鼠双侧卵巢,将去卵巢大鼠随机分为模型组、阳性药结合雌激素组、壮骨伸筋胶囊高、中、低剂量组(5.42,2.71,1.36 g·kg~(-1)),每组12只。连续ig 13周,每周连续ig 6 d,实验结束,取血和骨组织,测定骨密度及血清中血钙(Ca),碱性磷酸酶(ALP),抗酒石酸酸性磷酸酶(TRAP),雌二醇(E2),骨钙素(BGP),降钙素(CT)的变化;甲苯胺蓝染色观察骨组织病理学。结果:与假手术组比较,模型组大鼠骨密度,血清E2含量明显降低,大鼠血清ALP,TRAP,BGP含量明显升高(P<0.05,P<0.01),子宫系数明显降低(P<0.01),骨组织病理学观察显示模型组骨小梁稀疏,骨小梁间有断裂并且分离,间隙增大,排列紊乱,骨板层结构疏松,骨质疏松特征明显;与模型组比较,壮骨伸筋胶囊高、中剂量组能够显著增加模型大鼠的骨密度(P<0.01),增加模型鼠血清E2含量(P<0.05),降低模型大鼠血清ALP,TRAP,BGP含量(P<0.05),但是对血钙和CT作用却不明显,骨组织病理学观察显示骨组织结构趋于完整。结论:壮骨伸筋胶囊能提高去卵巢骨质疏松大鼠骨密度,提高雌激素水平,改善骨代谢水平,从而起到防治骨质疏松的作用。
Objective: To study the effect of Zhuanggu Shenjin capsules (ZSJ) on bone mineral density (BMD) and bone metabolism in ovariectomized rats and to explore its related mechanism. Methods: Seventy-two SD rats were randomly divided into sham operation group (n = 12) and model group (n = 60). The ovariectomized rats were randomly divided into model group Drug-conjugated estrogen group, Zhuanggunshenjian capsule high, medium and low dose group (5.42,2.71,1.36 g · kg -1), 12 in each group. Six weeks after continuous ig administration for 6 weeks, the rats were sacrificed and blood and bone tissues were harvested for measurement of bone mineral density (BMD), serum Ca, ALP, TRAP, Estradiol (E2), BGP and CT were measured. Tissue histopathology was observed by toluidine blue staining. Results: Compared with the sham operation group, the bone mineral density and serum E2 in the model group decreased significantly, while the contents of ALP, TRAP and BGP in the model group were significantly increased (P <0.05, P <0.01) 0.01). The bone histopathology showed that the trabecular bone was sparse and the trabecular bone was broken and segmented, the gap increased, the disorder was arranged, the structure of the osteoporosis was loose and the characteristics of osteoporosis were obvious. Compared with the model group, Compared with the model group, the bone mineral density (P <0.01), the serum E2 level (P <0.05) and the serum ALP, TRAP and BGP contents in model rats decreased significantly (P < 0.05), but the effect on serum calcium and CT was not obvious. The histopathological observation showed that the structure of bone tissue tended to be complete. Conclusion: Zhuanggu Shenjin Capsule can improve bone mineral density, increase estrogen level and improve bone metabolism in ovariectomized rats, which play a role in prevention and treatment of osteoporosis.
