目的研究胰腺腺癌细胞系 pRb- p16细胞调节途径相关因子的异常情况。方法用 PCR及单链构象多态性 (SSCP)的方法检测 5株人胰腺腺癌细胞系 p16基因的缺失和突变;用 Southern印迹分析 cyclin D1基因的扩增;用 Western印迹分析检测 pRb基因的表达。结果 2株胰腺腺癌细胞系存在 p16基因第 1外显子纯合性缺失,未发现第 1、 2外显子的基因突变; 1株细胞系有 cyclin D1基因的扩增; pRb基因在 5株细胞系中均呈高磷酸化的表达。结论胰腺腺癌细胞系中存在 pRb- p16细胞调节途径的异常,主要表现为 p16和 cyclin D1基因的改变。
Objective To investigate the abnormalities of the related factors in the regulatory pathway of pRb-p16 in pancreatic adenocarcinoma cell line. Methods The deletion and mutation of p16 gene in 5 human pancreatic adenocarcinoma cell lines were detected by PCR and single-strand conformation polymorphism (SSCP). The cyclin D1 gene was amplified by Southern blot analysis and the expression of pRb gene expression. Results There was homozygous deletion of exon 1 of p16 gene in two pancreatic adenocarcinoma cell lines and no mutation of exon 1 and exon 2 was found. One cell line had cyclin D1 gene amplification. The expression of pRb gene in 5 Strain cells were hyperphosphorylated expression. Conclusion The abnormalities of pRb-p16 cell line regulation exist in pancreatic adenocarcinoma cell lines, mainly showing the changes of p16 and cyclin D1 genes.
