目的探讨芦荟多糖对局灶性脑缺血大鼠的神经保护作用机制。方法采用线栓法阻断大鼠一侧大脑中动脉(MCAO),制备局灶性脑缺血模型,观察芦荟多糖对MCAO模型大鼠脑缺血后神经功能缺损导致的行为变化,用免疫组化法检测大脑皮质细胞凋亡相关蛋白Caspase-3的表达。结果芦荟多糖能抑制MCAO模型大鼠大脑皮质Caspase-3蛋白的表达。结论芦荟多糖对大鼠局灶性脑缺血具有保护作用,其机理与其影响、抑制Caspase-3的表达有关。
Objective To investigate the neuroprotective mechanism of aloe polysaccharides in rats with focal cerebral ischemia. Methods The cerebral artery occlusion (MCAO) of one side of the rat was blocked by a wire embolism method. Focal cerebral ischemia model was established to observe the behavioral changes caused by neurological deficits after cerebral ischemia in MCAO model rats. The expression of apoptosis-related protein Caspase-3 in cerebral cortical cells was detected by chemical method. Results Aloe polysaccharides inhibited the expression of Caspase-3 protein in cerebral cortex of MCAO rats. Conclusion Aloe polysaccharide has a protective effect on focal cerebral ischemia in rats. Its mechanism is related to its effect and inhibition of Caspase-3 expression.
