[目的]探讨焦炉工人DNA损伤与组蛋白H2A单泛素化水平的关系。[方法]采用高效液相色谱法对焦化厂106名焦炉工人(接触组)和73名水处理厂工人(对照组)尿中1-羟基芘(1-hydroxypyrene,1-OHP)浓度进行测定,并采用彗星实验和蛋白质印迹法(Western blot)分别检测DNA损伤和组蛋白H2A单泛素化水平。[结果]接触组Olive尾矩(Olive tail moments,OTM)和尿1-OHP的均值分别为0.46和0.34μmol/molCr,均显著高于对照组(分别为0.37和0.21μmol/molCr,P<0.05);而接触组单泛素化组蛋白H2A相对表达水平的中位数为0.61,与对照组(0.91)相比为降低(P<0.05)。以OTM值为应变量,多元线性回归显示自变量尿1-OHP和单泛素化组蛋白H2A进入回归方程,决定系数R2为0.383。[结论]焦炉工人DNA损伤程度与尿1-OHP含量和单泛素化组蛋白H2A相对表达水平关系密切。
[Objective] To investigate the relationship between DNA damage and mono-ubiquitination of histone H2A in coke oven workers. [Method] The concentration of 1-hydroxypyrene (1-OHP) in urine of 106 coke oven workers (contact group) and 73 water treatment factory workers (control group) in coking plant was determined by high performance liquid chromatography. The DNA damage and histone H2A mono-ubiquitination levels were detected by comet assay and Western blot respectively. [Result] The average values of Olive tail moments (OTM) and urinary 1-OHP in the contact group were 0.46 and 0.34μmol / molC, which were significantly higher than those in the control group (0.37 and 0.21μmol / molCr respectively, P <0.05 ). The median relative expression level of mono-ubiquitinated histone H2A in contact group was 0.61, which was lower than that in control group (0.91) (P <0.05). Using OTM as a dependent variable, multivariate linear regression showed that the independent variable urinary 1-OHP and monoubiquitylated histone H2A entered the regression equation with a coefficient of determination R2 of 0.383. [Conclusion] The degree of DNA damage in coke oven workers is closely related to the urinary 1-OHP content and the relative expression level of mono-ubiquitinated histone H2A.
