[目的]研究G市管网末梢水中有机提取物对雄性大鼠生殖器官重量及生殖激素分泌水平的影响,探讨其雄性生殖毒性作用。[方法]利用固相萃取法提取G市某区管网末梢水中有机污染物。将40只SD雄性大鼠随机分为溶剂对照组(给予玉米油)、低剂量染毒组(3L/kg.d)、中剂量染毒组(15L/kg.d)、高剂量染毒组(75L/kg.d),每组10只。每天灌胃一次,连续染毒28d后处死动物,采集大鼠血清,采用直接化学发光法检测大鼠血清中促黄体生成素(luteinizing hormone,LH)、睾酮(testosterone,T)及雌二醇(E2)的分泌水平。[结果]各染毒组大鼠双侧睾丸、附睾的重量与对照组比较,差异无统计学意义(P>0.05)。随管网末梢水中有机提取物染毒剂量的增加,大鼠血清中LH呈下降趋势,但与溶剂对照组比较,差异无统计学意义(P>0.05)。血清中E2水平随染毒剂量增加明显升高,与对照组及低剂量组比较,中、高剂量染毒组的E2均升高(P<0.05,P<0.01)。随染毒剂量的增加,T分泌水平呈先升高后降低的趋势,但仅高剂量组的降低具统计学意义(P<0.05)。[结论]G市某区管网末梢水有机提取物中、高剂量暴露,可损伤雄性大鼠睾丸间质细胞,抑制睾酮分泌,并诱导E2合成增加,破坏生殖内分泌激素的相对平衡,导致生殖功能受损。
[Objective] The purpose of this study was to investigate the effect of organic extract from the peripheral water of G network on reproductive organ weight and reproductive hormone secretion in male rats and its male reproductive toxicity. [Method] The organic pollutants in the water of the pipe network in a district of G city were extracted by solid phase extraction. 40 SD male rats were randomly divided into solvent control group (given corn oil), low dose group (3L / kg.d), medium dose group (15L / kg.d), high dose group (75L / kg.d), each group of 10. The rats were sacrificed every day for 28 days and the serum was collected. The serum levels of luteinizing hormone (LH), testosterone (T) and estradiol E2) secretion level. [Result] The weight of bilateral testis and epididymis in rats in each exposure group was not significantly different from that in control group (P> 0.05). With the increase of the dose of organic extract in the water at the distal end of the pipe network, the LH level in the serum of the rats decreased, but there was no significant difference between the two groups (P> 0.05). The level of E2 in serum increased significantly with the increase of dose, and the levels of E2 in middle and high dose groups were significantly higher than those in control group and low dose group (P <0.05, P <0.01). With the increase of exposure dose, the level of T secretion first increased and then decreased, but only decreased in high dose group with statistical significance (P <0.05). [Conclusion] The medium and high doses of organic extract from the peripherals of pipe network in a certain district of G city could damage the testicular stromal cells of male rats, inhibit the secretion of testosterone, induce the increase of E2 synthesis, destroy the relative balance of reproductive endocrine hormones, Impaired function.
