目的 探讨人类白细胞抗原B27亚型基因与急性前葡萄膜炎(acute anterioruveitis,AAU)易感性的关系。 方法 对49例临床确诊的AAU患者,通过聚合酶链反应(polym erase chain reaction,PCR)特异性扩增技术检测B27;以B27阳性者的DNA为模板对其人类白细胞抗原HLAB的第二、三外显子片段扩增。采用DNA测序技术对扩增产物做基因序列分析,经计算机处理获得受检者HLAB27亚型的信息。 结果 受检者中29 例为B27 阳性,占5939% ,其中仅发现B2704(13例,占4400% )和B27052(16例,占5600% )二种亚型基因携带者。二者间多数临床表现差异不大,但携带B27052基因的AAU患者伴发强直性脊柱炎(ankylosing spondylitis,AS)的人数(7 人,占2424% )显著高于B2704 基因携带者(1 人,占374% )。 结论 B2704和B27052 单独的亚型特异性与AAU 无易感性关联;但B27052基因可能与并发AS有关。
Objective To investigate the relationship between the human leukocyte antigen B27 subtype and the susceptibility to acute anterior uveitis (AAU). Methods 49 cases of clinically diagnosed AAU patients were detected by polymerase chain reaction (PCR) specific amplification of B27; B27 positive DNA as a template for its human leukocyte antigen HLA B second , Three exon fragments amplified. Using DNA sequencing technology to do gene sequence analysis of the amplified product, the information of the subject HLAB27 subtype was obtained by computer processing. Results 29 of the subjects were positive for B27, accounting for 5939%. Only B2704 (13 cases, 4400%) and B27052 (16 cases, 5600%) were found to be carriers of the two subtypes . The majority of AAU patients with B27052 gene were significantly associated with carriers of ankylosing spondylitis (AS, 7 persons, 24.24%) than carriers of B2704 gene (1 People, accounting for 374%). Conclusion The subtype specificity of B2704 and B27052 is not associated with susceptibility to AAU. However, the B27052 gene may be associated with complicated AS.
