镰刀形红细胞是由于红血球内有脱氧镰刀形血红蛋白(HbS)的长纤维形成所致。决定HbS多聚体形成的关键氨基酸残基,对开发药物和镰刀形细胞贫血的基因治疗非常重要。作者用自行设计的Hb表达系统(pHE2)的大肠杆菌中分别表达了4种质粒,共产生4种重组Hb(rHb);rHbS(β6Glu→Val;rHbS→D-Ibadan(β6Glu→Val,β87Thr→Lys);rHbS→Chiapas(β6Glu→Val,α114 Pro→Arg)和rHbS-Chiapas-D-Ibadan(β6Glu→Val,α114 Pro→Arg,β87Thr→Lys)。其两个突变位点(α114 Pro→
Sickle-shaped erythrocytes are due to the formation of long fibrils of deoxy-hemoglobin (HbS) in red blood cells. Determining the key amino acid residues formed by HbS multimers is important for gene therapy for developing drugs and sickle cell anemia. Four recombinant plasmids (rHb) were produced in E. coli with a self-designed Hb expression system (pHE2). Four recombinant Hb (rHb) were produced.
